چکیده
Hexameric hemoprotein (HTHP) is employed as a scaffold protein for the supramolecular assembly and activation of apoptotic signalling enzyme caspase-9, using short DNA elements modular recruitment domains. Caspase-9 on HTHP platform due to enhanced proximity followed by combinatorial inhibition at high concentrations. The domains allow reversible switching caspase-9 activity state modulatory strands. Tuning domain affinity allows generating kinetically trapped active complexes, well dynamic repositioning caspases over populations monovalent sink platforms. conceptual combination highly structured multivalent with provides emergent biomimicry properties advanced levels control assembly.

Adiponutrin: a multimeric plasma protein.
The interest in adiponutrin stems from adiponutrin variant I148M, which is strongly associated to non-alcoholic fatty liver disease. Adiponutrin has to date been considered to be solely an intracellular protein, with a role in lipid metabolism in liver and adipose tissue. However, a physiologically relevant role for adiponutrin has not been found. The aim of this study was to investigate the pr...
متن کاملSynthetic mimetics of protein secondary structure domains
Proteins modulate the majority of all biological functions and are primarily composed of highly organized secondary structural elements such as helices, turns and sheets. Many of these functions are affected by a small number of key protein-protein contacts, often involving one or more of these well-defined structural elements. Given the ubiquitous nature of these protein recognition domains, t...
متن کاملDiscovering Domains Mediating Protein Interactions
Background: Protein-protein interactions do not provide any direct information regarding the domains within the proteins that mediate the interactions. The majority of proteins are multi domain proteins and the interaction between them is often defined by the pairs of their domains. Most of the former studies focus only on interacting domain pairs. However they do not consider the in...
متن کاملL 1 Protein kinase A : a dynamic and multivalent scaffold
cAMP-Dependent protein kinase has served as a prototype for the protein kinase superfamily in many ways. Its structures provide a comprehensive set of conformational states that reflect the different stages of catalysis, and recent holoenzyme structures have expanded our understanding of how PKA can recognize diverse proteins that dock to sites that are distal to the active site [1–4]. By compa...
متن کاملIncreased recruitment of TATA-binding protein to the promoter by transcriptional activation domains in vivo.
The rate at which the TATA-binding protein (TBP) interacts with the TATA element and promotes transcription by RNA polymerase II was determined in yeast cells. A TBP derivative with altered TATA-element specificity was rapidly induced, and transcription from promoters with appropriately mutated TATA elements was measured. Without a functional activator protein, basal transcription was observed ...
